Description: Some form of LPS is a major component of the cell membrane of Gram negative bacteria,contributing greatly to the structural integrity of the bacteria, and protecting the membranefrom certain kinds of chemical attack. LPS is an endotoxin, and induces a strong responsefrom normal animal immune systems. LPS function has been under experimental researchfor several years due to its role in activating many transcriptional factors, which becomeactive after stimulation with LPS. LPS also produces many types of mediators involved inseptic shock. The J5 mutant of E. coli lacks the enzyme uridine diphosphate glucose 4-epimerase andtherefore produces an incomplete LPS, deficient in galactose and all the sugars distal tothe central polymers. The 55 mutant has no O-specific chains and its endotoxin remains asthe core LPS containing lipid A, N acetyl glucosamine, 2-keto-3-deoxyoctonate, heptoseand glucose (a composition similar to that of the Rc strains of Salmonella).

Specificity: The antibody recognizes E. coli J5 LPS. It has also been found to react with K. pneumoniae, S. sonnei and S. typhimurium.

Application Details

Application Notes: Immunofluorescence. ELISA: (a 1 mg/ml solution has been used at dilutions of up to 1/1500). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Concentration: 0.5 mg/ml

Buffer: PBS, pH 7.2, containing 0.09% Sodium Azide.

Storage: Store the antibody undiluted at 2-8°C for one month or (in aliquots) at -20°C for longer. Avoid repeated freezing and thawing. Shelf life: one year from despatch.
