  • Specificity

This Monoclonal Antibody 2E1 recognizes the three isoforms of the Fc-gamma RII designated as A, B and C. Monocytes carry all forms of Fc-gamma RII, Neutrophils carry Fc-gamma RIIA and Fc-gamma RIIC. Fc-gamma RIIB is expressed by B lymphocytes and mast cells.

    Cross-Reactivity (Details)

    Species reactivity (tested):Human.

    Purification

    Affinity Chromatography

    Immunogen

    K562 cells.

    Isotype

    IgG2a
  • Application Notes

FACS: 10 μL stock solution to label 10^6 cells. Immunohistochemistry on Frozen Sections: 2 μg/mL (1/200). ELISA. Immunoprecipitation. Suggested Positive Control: Human tonsil.
    Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.

    Protocol

Protocol with frozen, ice-cold acetone-fixed sections: The whole procedure is performed at room temperature1. Wash in PBS2. Block endogenous peroxidase3. Wash in PBS4. Block with 10% normal goat serum in PBS for 30min. in a humid chamber5. Incubate with primary antibody (dilution see datasheet) for 1h in a humid chamber6. Wash in PBS7. Incubate with secondary antibody (peroxidase-conjugated goat anti mouse IgG+IgM(H+L) minimal-cross reaction to human) for 1h in a humid chamber8. Wash in PBS9. Incubate with AEC substrate (3-amino-9-ethylcarbazol) for 12min. 10. Wash in PBS11. Counterstain with Mayer's hemalum.

    Restrictions

    For Research Use only
  • Reconstitution

Restore with 0.5 mL distilled water (stock solution contains 0.4 mg/mL IgG)

    Concentration

    0.4 mg/mL

    Buffer

PBS, pH 7.2 containing 0.01 % Thiomerosal as preservative and 10 mg/mL BSA as stabilizer

    Handling Advice

Dilute only prior to immediate use

    Storage

    4 °C

    Storage Comment

Store the antibody at 2-8 °C before opening. DO NOT FREEZE! This product is stable at 2-8 °C as an undiluted liquid. For extended storage after reconstitution, add glycerol to a final concentration of 50% and store at -20 °C. Freezing alkaline phosphatase conjugates without glycerol will result in a substantial loss of enzymatic activity.
  • Target

FCGR2A (Fc Fragment of IgG, Low Affinity IIa, Receptor (CD32) (FCGR2A))

    Alternative Name

    CD32 / FcRII-A

    Background

CD32 is a receptor for the Fc region of complexed or aggregated immunoglobulins gamma. This is a low affinity receptor. It is Involved in a variety of effector and regulatory functions such as phagocytosis of immune complexes and modulation of antibody production by B cells. Binding to this receptor results in downmodulation of previous state of cell activation triggered via antigen receptors on B cells (BCR), T cells (TCR) or via another Fc receptor.Synonyms: CDw32, FCG2, FCGR2A, FCGR2A1, Fc-gamma RII-a, Fc-gamma-RIIa, IGFR2, IgG Fc receptor II, Low affinity immunoglobulin gamma Fc region receptor II
