  • Application Notes

    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofh-IFN-gamma (50 pg/mL), a concentration of 0.04-0.06 μg/mL of the previous lot of thisantibody was required. ELISA: To detect h-IFN-gamma by direct ELISA (using 100 l/well antibody solution) aconcentration of at least 0.5 μg/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with compatible secondary reagents, allows the detectionof 0.2-0.4 ng/well of recombinant h-IFN-gamma. Western blot: To detect h-IFN-gamma by Western Blot analysis this antibody can be used ata concentration of 0.1- 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant h-IFN-gamma is 1.5-3.0 ng/lane, under either reducingor non-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Interferon gamma is mainly produced by T lymphocytes and NK cells. It is a pleiotropic cytokine involved in the regulation of nearly all phases of immune and inflammatory responses,including the activation, growth and differentiation of T cell, B cells, macrophages, NK cells and other cell types such as endothelial cells and fibroblasts. It has weak antiviral and antiproliferative activity, and poteniates the antiviral and anti tumor effects of IFN alpha / beta (type I interferon). It is upregulated by IL2, FGF basic, EGF and downregulated by vitamin D3 or DMN. Labile at pH 2.Synonyms: IFN-gamma, gamma IFN
