Application Notes

ELISA. Western blotting: 0.5 m g/mL. Sample preparation: Resuspend approx. 50 mil. cells in 1 mLcold Lysis buffer (1 % laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8.2, 50 mM NaF including Protease inhibitor Cocktail). Incubate 60 min on ice. Centrifuge to remove cell debris. Mix lysate with reducing Laemmli SDS-PAGE sample buffer. Reducing conditions. Functional Assay. Antibody ABIN120632 strongly inhibits the enzyme activity of HIV-1 and HIV-2 proteases.

Background
- The HIV protease (PR) hydrolyzes polyproteins of HIV virus into functional protein products that are essential for its assembly and subsequent activity. This maturation process occurs as the virion buds from the host cell.
