Description Chain (34-kDa) of chicken CD8 In the chicken, the CD8 molecule is present in two forms: (i) a homodimer of two chains, and (ii) a heterodimer of an chain and a chain.1 While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8- and CD8-chains, a relatively large proportion of the CD8+ TCR- cells in the spleens of embryos and young chicks express only the -chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved, but there is a population of TCR- CD8 cells in the chicken that is not found in rodents.1,3 Chicken CD8 is expressed on approximately 80% of thymocytes, 15% of blood mononuclear cells and 50% of spleen cells, but <1% of cells in the bursa and bone marrow.1,2 MAb CT-8 recognizes the CD8 chain.

Application Notes Working Dilutions: Flow Cytometry: Fluorescein (1 ug/10^6 cells), Biotin (1 ug/10^6 cells), R-phycoerythrin (0.2 ug/10^6 cells), Cyanine 5 (1 ug/10^6 cells). Since applications vary, you should determine the optimum working dilution of the product that is appropriate for your specific need.
Storage The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.0. No preservatives or amine-containing buffer salts added. Store at 2-8C. The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8C. Do not freeze! Protect conjugated forms from light. Each reagent is stable for the period shown on the bottle label if stored as directed. The Cyanine 5 (CY5) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed. WARNING Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water.
