Background
- HOX11, also called homeobox11 or Tcl-3 is located at the 10q24 T-Cell translocation breakpoint region. It encodes a homeobox-domain containing protein, HOX11, containing a glycine and proline-rich amino terminus. HOX11 is required for maintenance of the developing spleen and cell survival. The HOX11protein binds to a specific DNA sequence and localizes to the cell nucleus. It transactivates transcription of a reporter gene linked to a cis-regulatory element, acting as a positive transcription activator. The HOX11 gene is highly expressed in T-ALLs as a result of a t (7:10) (q35,q24) or t(10:14)(q24,qll) translocation. The homeobox gene is deregulated upon translocation into the T-cell receptor locus and activates a specific subset of tumor-associated target genes.
