    Background
    Phospholamban (PLB/PLN) is a 52 amino acid phosphoprotein which regulates the calcium pump of cardiac sarcoplasmic reticulum (SR). PLB is an oligomer of five identical subunits each having a cytoplasmic and transmembrane domain. The cytoplasmic domain (residues 1 to 25) contains cAMP-dependent protein kinase phosphorylation sites and is highly basic and readily cleaved by proteases, whereas the transmembrane domain (residues 25 to 52) is mostly hydrophobic, protease resistant and stabilizes the pentamer. PLB is an inhibitor of cardiac muscle sarcoplasmic reticulum Ca(2+)-ATPase in the unphosphorylated state, but inhibition is relieved upon phosphorylation of the protein. The subsequent activation of the Ca(2+) pump leads to enhanced muscle relaxation rates, thereby contributing to the inotropic response elicited in heart by beta-agonists. Mutations in this gene are a cause of inherited human dilated cardiomyopathy with refractory congestive heart failure.Synonyms: Cardiac phospholamban, PLB, PLN
