Description
|
Cytokeratins are a subfamily of intermediate filaments and characterized by remarkablebiochemical diversity. Cytokeratins are represented in epithelial tissues by at least 20different polypeptides, molecular weight between 40 kDa and 68 kDa. The individualcytokeratin polypeptides are designated 1 to 20 and divided into the type I (acidiccytokeratins 9-20) and type II (basic to neutral cytokeratins 1-8) families.Cytokeratins 18 belongs to type I family (acidic cytokeratins).

|
Western Blotting - Recommended dilution: 1-2 µg/ml, overnight in 4 o C Positive control: HeLa human cervix carcinoma cell lineMCF-7 human breast adenocarcinoma cell line. Sample preparation: Resuspend approx. 50 mil. cells in 1 ml cold Lysis buffer (1%laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8. 2, 50 mM NaF including Proteaseinhibitor Cocktail). Incubate 60 min on ice. Centrifuge to remove cell debris. Mix lysate withnon-reducing SDS-PAGE sample buffer. Application note: Non-reducing conditions. SDS-PAGE (10% separating gel). ImmunocytochemistryELISA. Application note: The antibody C-04 has been tested as the capture antibody in a sandwichELISA for analysis of Cytokeratin 18 in combination with antibody DA-7 (cat. no. 1B-110-C100) and with the antibody DC-10 (cat. no. 1B-107-C100).
