Additional Information

Characteristics For quantitative detection of mouse CXCL1 in sera, plasma, body fluids, tissue lysates or cell culture supernates.
Alternative name C-X-C Motif Chemokine 1 (GRO Alpha) (CXCL1)/GROalpha
Description Chemokine (C-X-C motif) ligand 1 (CXCL1) is a small cytokine belonging to the CXC chemokine family that was previously called GRO1 oncogene, GROa, KC, Neutrophil-activating protein 3 (NAP-3) and melanoma growth stimulating activity, alpha (MSGA-a). In humans, this protein is encoded by the CXCL1 gene. The gene for CXCL1 is located on human chromosome 4 amongst genes for other CXC chemokines. The mature form of CXCL1 is maximally 73 amino acids long. CXCL1 is secreted by human melanoma cells, has mitogenic properties and is implicated in melanoma pathogenesis. 1 CXCL1 is expressed by macrophages, neutrophils and epithelial cells, 2 and has neutrophil chemoattractant activity. 3 This chemokine elicits its effects by signaling through the chemokine receptor CXCR2.4 CXCL1 decreased the severity of multiple sclerosis and may offer a neuro-protective function.5 The standard product used in this kit is recombinant mouse CXCL1, consisting of 77 amino acids with the molecular mass of 8KDa.
Specificity No detectable cross-reactivity with any other cytokine.
Sensitivity Sensitivity: < 1pg/ml. Range: 15.6pg/ml-1000pg/ml

Application Details

Principle This mouse CXCL1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. Mouse CXCL1 specific-specific monoclonal antibodies (clone No. 48415) were precoated onto 96-well plates. The mouse specific detection polyclonal antibodies were biotinylated. The test samples and biotinylated detection antibodies were added to the wells subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CXCL1 amount of sample captured in plate.
Application Notes Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing. Don
Components Lyophilized recombinant mouse CXCL1 standard: 10ng/tube
