Principle

The DetectX® 13,14-dihydro-15-keto-PGF2a (PGFM) Immunoassay kit is a patent pending assayfrom Dr. Martin Dehnhard at Leibniz Institute for Zoo & Wildlife Research, Berlin, Germany licensedexclusively and designed to quantitatively measure PGFM present in fecal extracts, urine, serumand plasma samples. Please read the complete kit insert before performing this assay. A PGFMstandard is provided to generate a standard curve for the assay and all samples should be read offthe standard curve. Standards or diluted samples are pipetted into a clear microtiter plate coatedwith an antibody to capture rabbit IgG. A PGFM-peroxidase conjugate is added to the standardsand samples in the wells. The binding reaction is initiated by the addition of a rabbit polyclonalantibody highly specific to PGFM to each well. After an hour incubation, the plate is washed andsubstrate is added. The substrate reacts with the bound PGFM-peroxidase conjugate. After ashort incubation, the reaction is stopped and the intensity of the generated color is detected in amicrotiter plate reader capable of measuring 450 nm wavelength. The concentration of the PGFMin the sample is calculated, after making suitable correction for the dilution of the sample, usingsoftware available with most plate readers.
