Plasticity allows Arl1 to interact with different effectors of unrelated structure.
Together the findings indicate that Arl1 links Rab4 (show RAB4A Antibodies)-dependent formation of endosomal sorting domains with downstream assembly of adaptor protein complexes that constitute the endosomal sorting machinery.
The Arl1.Arfaptin-2 (show ARFIP2 Antibodies) BAR structure suggests that one of the two Arl1 molecules competes with Rac1, which binds to the concave face of the Arfaptin-2 (show ARFIP2 Antibodies) BAR homodimer and may hinder its membrane association.
arfaptins are recruited onto trans-Golgi membranes by interacting with Arl1, and capable of inducing membrane deformation via their BAR domains.
Data show that Arl1 regulates the membrane recruitment of Golgin-97 (show GOLGA1 Antibodies), which plays a role in transport from the endosome to the trans-Golgi network.
These data suggest that lethality of Arfrp1 (show ARFRP1 Antibodies) knockout embryos is due to a specific disruption of protein targeting, e.g., of ARL1 and Golgin-245 (show GOLGA4 Antibodies), to the Golgi.
Results show that ARL1 and ARFRP1 (show ARFRP1 Antibodies) regulate retrograde transport of Shiga toxin to the TGN (show TG Antibodies) and anterograde transport of VSVG from the TGN (show TG Antibodies), respectively.
Study demonstrates that Golgi recruitment of endogenous GCC185 (show GCC2 Antibodies) does not involve Rab6A (show RAB6A Antibodies)/A' and Arl1.
These findings suggest that ARF-1.1 interacts with NCS-1 (show NCS1 Antibodies) in AIY neurons and potentially pifk-1 in the Ca(2 (show CA2 Antibodies)+) signaling pathway that leads to inhibited locomotion at an elevated temperature.
Arl1 does not appear to be required for AP-1 recruitment in all tissues, suggesting that it is crucially required to enhance Arf1 (show ARF1 Antibodies) activation at the trans-Golgi in particular tissues.
The protein encoded by this gene belongs to the ARL (ADP-ribosylation factor-like) family of proteins, which are structurally related to ADP-ribosylation factors (ARFs). ARFs, described as activators of cholera toxin (CT) ADP-ribosyltransferase activity, regulate intracellular vesicular membrane trafficking, and stimulate a phospholipase D (PLD) isoform. Although, ARL proteins were initially thought not to activate CT or PLD, later work showed that they are weak stimulators of PLD and CT in a phospholipid dependent manner.
