- Background
-
GFRA3, GFRα3, GDNF family receptor α3, GDNF receptor alpha 3,GFRα3 (GDNF Family Receptor α3) belongs to the glial cell line derived neurotrophic factor (GDNF) family of receptors which contains four members GFRα1-4. The GDNF family of ligands (GFLs)- GDNF, Neurturin, Artemin, and Persephin bind to co-receptors GFRα1-4 and activate Ret receptor tyrosine kinase. GDNF-family receptor α is glycosyl-phosphatidylinositol anchored to the cell surface. GFRα3 structure contains three homologous domains (D1, D2 and D3) and a C-terminal extension1-4.GFRα3 is a highly selective receptor for Artemin (Artn). Artemin is a vasculature-derived neurotrophic factor that modulates the development and function of sensory neurons and participates in the pathophysiology of peripheral inflammation and pain hyperalgesia. In adults, GFRα3 is restricted to neurons of the peripheral nervous system. The protein is expressed by a subpopulation of nociceptive sensory neurons which also express the Ret receptor tyrosine kinase, TRPV1, TRPA11,2,3.
