- Hydroxymethylated DNA immunoprecipitation (hmeDIP)
- The hMeDIP Kit is tested for specific enrichment of 5-hydroxymethylcytosine DNA as compared with 5-methylcytosine DNA or unmethylated DNA.
- The hMeDIP Kit is designed to immunoprecipitate and enrich for DNA fragments containing 5-hydroxymethylcytosine (5-hmC). The hMeDIP Kit contains a highly specific purified 5-hmC antibody and the necessary buffers to perform methylated DNA immunoprecipitation (MeDIP). The affinity of the antibody enables separation of 5-hmC DNA from 5-mC DNA. Additionally, the antibody works to efficiently immunoprecipitate either double-stranded or single-stranded input DNA.
The hMeDIP kit is run on 500 ng of MseI digested human genomic DNA that has been spiked with 25 pg of control DNA and assayed in real time PCR with the APC PCR primer mix included in the hMeDIP kit. The human genomic DNA used in the IP reaction is not methylated at the APC locus and therefore the only DNA recovered in the control DNA reactions will be from the spike. The specificity of the hMeDIP kit to specifically enrich for 5-hydroxymethylcytidine-containing DNA fragments is shown in Figure 1.
