- Resuspend cells in assay media free of h-GM-CSF containing 10 % FBS and 2X Gentamicin. Transfer 100ml culture/well to assay plate (19,500 cells/well, passage # 13). Serial dilute h-GM-CSF in assay media in duplicate. Transfer 100 ml/well serial diluted h-GM-CSF to cells in assay plate. Final assay volume of each well is 200 ml containing 2X Gentamicin and h-GM-CSF. Incubate 70 hours. Add 20 ml of Promega Substrate Cell Titer 96 Aqueous One Solution Reagent to each well. Incubate 37°C and read OD at 490 nm. The ED50 for this effect is 0.06 - 0.09 ng/ml.

- Background
- GM-CSF is a hematopoietic growth factor that stimulates the development of neutrophils and macrophages and promotes the proliferation and development of early erythroid megakaryocytic and eosinophilic progenitor cells. It is produced in by endothelial cells, monocytes, fibroblasts and T-lymphocytes. GM-CSF inhibits neutrophil migration and enhances the functional activity of the mature end-cells. Synonym: rHuGM-CSF 7, hGM-CS. Formulation: Lyophilized from a 0.2 mym filtered solution in 10 mM PB, pH7.0.
