Immunogen: The whole rabbit serum was prepared by repeated immunizations with recombinant human IL-10 produced in E.coli.

Specificity: This antiserum has been heated to 56oC for 30 minutes. The antiserum detects recombinant and native IL-10 present in body fluids and cell supernatants in various assays (ie. IL-1 stimulated IL-10 production from fibroblasts). In Western blot analysis of natural cell products or human body fluids, multiple bands of IL-10 will appear due to the variable amount of glycosylation on the molecule. The antiserum is also useful for neutralization of human of IL-10 activity in bioassays. For neutralization, incubate the sample with a 1:400 dilution of the antiserum for at least 4 hours before being tested. A control of similarly diluted normal rabbit IgG (heat inactivated) is recommended. The antibody will not neutralize the biological activity of murine IL-10. Application Notes: Note: This antiserum has been tested for use in neutralizations, ELISA, immunohistochemistry, radioimmunoassay, immunoprecipitation, immunohistochemistry and immunoblotting. The antibody recognizes hrIL-10. Reactivity in other immunoassays is unknown. Recommended Dilutions : This product was assayed by immunoblot using HRP Goat-anti-Rabbit IgG [H&L] (code ABIN101990) and TMB as a substrate. A working dilution range of 1:400 to 1:800 is suggested for this application to detect IL-10 from supernatants or lysates of 2 x 10 6 endotoxin-stimulated human peripheral blood mononuclear cells (PBMC). PBMC are stimulated for 24 hours with 1% (v/v) human serum plus 10 ng/mL E.coli LPS. A dilution range of 1:400 is suggested for use in immunohistochemistry using either paraffin fixation or cryofixation. This product has been assayed for the ability to immunoprecipitate antigen. A dilution range of 1:400 to 1:800 is suggested for this immunoassay. For immunoprecipitation, pre-clearing with a non-specific rabbit IgG is helpful to reduce background. This product has been assayed by ELISA against IL-10 using HRP Conjugated Anti-Rabbit IgG [H&L] (Goat) (code ABIN101990) and ABTS as a substrate for 30 minutes at room temperature. A working dilution range of 1:400 to 1:1,200 is suggested for this product. For use in ELISA formats, this antibody is best used as the second antibody in combination with a monoclonal antibody as a capture antibody. By ELISA this antibody shows less than 1% cross-reactivity with murine IL-10. Direct ELISA results in no cross-reactivity against other cytokines. This product has been assayed by radioimmunoassay against antigen. A dilution of 1:4,000 is suggested for this immunoassay. Researchers should determine optimal titers for other applications.
