Description
|
The SET domain is a highly conserved,approximately 150-amino acid motif implicated in the modulation of chromatin structure. Itwas originally identified as part of a largerconserved region present in the Drosophila trithorax protein and was subsequentlyidentified in the Drosophila Su(var)3-9 and 'Enhancer of zeste' proteins, from which theacronym SET isderived. Studies have suggested that the SET domain may be a signature of proteins thatmodulate transcriptionally active or repressed chromatin states through chromatinremodeling activities. ESET functions as a histone methyltransferase by methylation ofLys-9 of histone H3. H3 Lys-9 methylation represents a specific tag for epigenetictranscriptional repression by recruiting HP1 proteins to methylated histones. ESET shows anuclear localization and is associated with non-pericentromeric regions of chromatin, andis excluded from nucleoli and islands ofcondensed chromatin. Although ESET is a widely expressed protein, it is highly expressedin the testis.

Application Notes
|
This affinity purified antibody has been tested for use in ELISA (1: 10,000-1: 50,000) andWestern blotting (1: 1,000-1: 2,500). Western blotting shows reactivity specific for human ESET detecting a band atapproximately 170 kDa. Specific band detection by Western blot is blocked by peptidecompetition by pre-incubating the antibody with the immunizing peptide prior to reactionwith the membrane. Reactivity in other immunoassays is unknown. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
