Application Details

Application Notes: Immunohistochemistry on paraffin embedded sections: 16 μg/mL. Antigen Retrieval: Steam slides in 0.01 M sodium citrate buffer, pH 6.0, at 99-100 °C for 20 min. Remove from heat and let stand at room temperature in buffer for 20 min. Rinse in 1x TBS with Tween (TBST) for 1 min. at room temperature.
Other applications not tested.

Background: CXCR3, a Chemokine Receptor, is highly similar to the interleukine 8B receptor and has been shown to mediate recruitment of T cells to sites of inflammation and retention of T-cells in the inflamed CNS. CXCR3 has been suggested to be the principal inflammatory chemokine receptor involved in intrathecal accumulation of T-cells in multiple sclerosis and diabetes. It is expressed on malignant B cells in chronic lymphoproliferative disorders (Jones et al., 2000). Ligand: Interferon gamma-inducible protein 9, Interferon gamma-inducible protein 10 and Interferon gamma inducible CXC chemokines I-TAC, IP10 and Mig. An alternatively spliced isoform, CXCR3B, has recently been identified (Lasagni et al. 2003).
