Description The MDM2 onco-protein is overexpressed in human cancer cells and itpromotes tumorigenesis by directly blocking thefunction of the p53 tumor suppressor. Studiesalso indicate that MDM2 regulates cell prolifer-ation via p53 independent mechanisms. MTBP(Mdm2 [Two] Binding Protein) was identified bya yeast two-hybrid screen as an MDM2 bindingprotein (1). MTBP has been reported tomodulate cell cycle progression by a p53independent mechanism and suppress MDM2-mediated p53 degradation when ectopicallyexpressed in cells (1,2). The human MTBPgene localizes to 8q24 (3) and MTBP RNA isexpressed in a wide range of tissues, withhighest being in the thymus, testes and ovaries(1).
Specificity This antibody is directed against human MTBP protein. The product was producedfrom clarified mouse ascetic fluid. A BLAST analysis was used to suggest reactivity with this protein fromhuman (100% homology) and chimpanzee. Cross reactivity with MTBP protein from mouse (77% homology)and chicken (79% homology) is unlikely due to the relatively low number of positive residues and high numberof gaps within this sequence.
