Description
|
Murine CD107a/LAMP-1 (Mr 110-140 kDa with a core protein of Mr ~40 kDa) CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated, type I transmembrane protein that constitutes one of the two major sialoglycoproteins on lysosomal membranes that can be used to distinguish lysosomes from other organelles.1 It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion.2 CD107a is principally expressed in epithelial cells and macrophages in a variety of organs in normal and Beige mutant mice.3 LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases.

Application Notes

Working Dilutions: Flow Cytometry: Purified (1 ug/10^6 cells), Fluorescein (1 ug/10^6 cells), Allophycocyanin (0.2 ug/10^6 cells). Since applications vary, each investigator must determine the optimum working dilutions of the product that is appropriate for their specific needs.

Storage
|
The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.0. No preservatives or amine-containing buffer salts added. Store at 2-8C. The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The allophycocyanin (APC) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8C. Do not freeze! Protect conjugated forms from light. Each reagent is stable for the period shown on the bottle label if stored as directed. WARNING Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
