Application Notes: Peptide ELISA: antibody detection limit dilution 1:32,000. Western Blot: Preliminary experiments gave an approx 26kDa band in lysates of Human Brain, of T cell line MOLT4, of myeloid erythroblast cell line K562 and of EBV-immortilized lymphoblastoid after 1µg/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 179kDa according to NP_002326.1. The 26kDa band was successfully blocked by incubation with the immunizing peptide. We would appreciate any feedback from people in the field - have any results been reported with other antibodies/lysates? Have any further splice variants/modified forms been reported? Immunohistochemistry, full caption: In paraffin embedded Human Pancreas shows staining of cytoplasm in some cells of the islets of Langerhans. Recommended concentration, 1-2µg/ml. Microwaved antigen retrieval with Tris/EDTA buffer pH9, HRP-staining. Western Blot: ABIN185353 has been successfully used in WB assays with rabbit tissue lysate (whole bone, amount loaded: 20µg, antibody dilution 1:200, incubation time of primary antibody: 2 hours, milk (5%) as blocking agent for 1.5 hours, sewcondary antibody: bovine anti-goat with incubation time of 2 hours). Rabbit Bone samples were used and probed at the same time with GAPDH as a loading control. Specific bands of 51kDa (LRP5) and 37kDa (GAPDH) were detected. Antibody buffer was 1% NFDM in TBS. (Information was provided by a customer).

Purification: Purified from goat serum by ammonium sulphate precipitation followed by antigen affinity chromatography using the immunizing peptide.

Buffer: 0.5 mg/ml in Tris saline, 0.02% sodium azide, pH7.3 with 0.5% bovine serum albumin.

Storage: Aliquot and store at -20°C. Minimize freezing and thawing.
