Description The CD8 T cell coreceptor (monomer approx. 32-34 kDa) is expressed as alpha/betaheterodimer on majority of MHC I-restricted conventional T cells and thymocytes and asalpha/alpha homodimer on subsets of memory T cells, intraepithelial lymphocytes, NK cellsand dendritic cells. Regulation of CD8 beta level on T cell surface seems to be an importantmechanism to control their effector function. Assembly of CD8 alpha-beta but notalpha-alpha dimers is connected with formation or localization to the lipid rafts. Recruitingtriggered TCR complexes to these membrane microdomains as well as affinity of TCR toMHC I is modulated by CD8, thereby affecting the functional diversity of the TCR signaling.
Specificity The antibody MEM-31 recognizes a conformationally-dependent epitope of CD8, a cellsurface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cellinteractions within the immune system. CD8 is a disulfide-linked dimer and exists as a CD8alpha/alpha homodimer or CD8 alpha/beta heterodimer (each monomer approx. 32-34 kDa).The antibody does not react with formaldehyde-fixed cells, negative in WesternBlotting application.HLDA III, WS Code T 575

Application Details

Application Notes The reagent is designed for Flow Cytometry analysis of human blood cells using 10 µl reagent / 100 µl of whole blood or 10 6 cells in a suspension. The content of a vial (1 ml) is sufficient for 100 tests.
Buffer The reagent is provided in phosphate buffered saline (PBS) containing 15 mM sodium azideand 0.2% (w/v) high-grade protease free Bovine Serum Albumin (BSA) as a stabilizing agent.
Storage Store in the dark at 2-8 o C. Do not freeze. Avoid prolonged exposure to light. Do not use after expiration date stamped on vial label.Short-term exposure to room temperature should not affect the quality of the reagent.However, if reagent is stored under any conditions other than those specified, the conditions must be verified by the user.
