Description SHIP-1 (SH2 domain containing inositol phosphatase-1) is a 5'inositol phosphatase thatregulates cell responses in lymphocytes and myeloid cells by hydrolyzing the secondmessenger PI(3,4,5) trisphosphate. SHIP-1 is recruited upon engagement of both inhibitoryand activatory receptors, such as FcgammaRIIB, Fcgamma RIII, FcepsilonRI or cytokineand growth factor receptors, and supresses PI3K-dependent signaling, down-regulates cellmigration and invasion of transformed cells and phagocytosis. SHIP-1 also serves as ascaffold for the recruitment of other proteins to the plasma membrane.
Specificity The antibody SHIP-02 reacts with SHIP-1, a phosphoinositide phosphatase largely confinedto hematopoietic cells. Multiple forms of SHIP-1 have been reported with molecular weightsof 110, 125, 130, 135 and 145 kDa.

Application Details

Application Notes Flow Cytometry: Recommended dilution: 2-5 µg/ml. Positive control: human blood leukocytes. Application note: - Positive control: RAMOS human cell line. Sample preparation: Resuspend approx. 50 mil. cells in 1 ml cold Lysis buffer (1%laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8. 2, 50 mM NaF including Proteaseinhibitor Cocktail). Incubate 60 min on ice. Centrifuge to remove cell debris. Mix lysate withreducing Laemmli SDS-PAGE sample buffer. Boil for 6 min. Application note: Reducing conditions.
