Specificity

Application Notes
- Immunohistochemistry (IHC) 1:100-1:400 for 30 minute at room temp. using polymer system or streptavidin-biotin system. Formalin-fixed paraffin-embedded (FFPE) tissue sections and cell smears require antigen retriever, boiling tissue in 10 mM citrate buffer, pH 6.0 for 10-20 minute, followed by cooling at room temp. for 20 minutes. The optimum dilution should be determined by the individual lab. Positive Control: Human kidney. Cellular Localization: Connective tissue matrix and basement membrane.

Background
- Fibronectin is a glycoprotein present in a soluble dimeric form in plasma, and in a dimeric or multimeric form at the cell surface and extracellular matrix. Fibronectin is involved in cell adhesion and migration process including embryogenesis, wound healing, blood coagulation, host defense, and metastasis. They also occur as dimmer of two 200 kDa subunits. Fibronectin has been implicated in carcinoma development in lung cancer. Fibronectin expression is increased especially in non-small cell lung carcinoma. The adhesion of lung carcinoma cells to Fibronectin enhances tumorgenecity and confers resistance to apoptosis induced by standard chemotherapeutic agents.
