Description P-selectin, also called GMP-140, CD62, or selectin P, is a 140-kD adhesion molecule, expressed at the surface of activated cells, that mediates the interaction of activated endothelial cells or platelets with leukocytes.1 It is stored in secretory granules and expressed at the plasma membrane after cell activation.2 It is known to play an important role in atherosclerosis.3 The major ligand for P-selectin on leukocytes is P-selectin glycoprotein ligand-1 (PSGL-1).4 The standard product used in this kit is recombinant mouse P-Selectin, excluding intercellular P-Selectin and transmembrane domain. It has 42-709 amino acids sequence with the molecular mass of 99.1KDa. As a result of glycosylation, the molecular mass of 190-200KDa is revealed by SDS-PAGE.
Specificity No detectable cross-reactivity with any other cytokine.
Sensitivity Sensitivity: < 5 pg/ml. Range: 62.5pg/ml-4000pg/ml

Application Details

Principle This mouse P-Selectin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. Mouse P-Selectin specific-specific polyclonal antibodies were precoated onto 96-well plates. The mouse specific detection polyclonal antibodies were biotinylated. The test samples and biotinylated detection antibodies were added to the wells subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse P-Selectin amount of sample captured in plate.
