Mouse Goat Un-conjugated IP, ELISA HeLa lysate either in RIPA buffer (lanes 1, 2, 4, 6) or TX100 buffer (lanes 3, 5, 7) overexpressing Mouse CSTF2 (64 kDa)  (DYKDDDDK tagged) immunoprecipitated using  ABIN1686783 (2 µg)  with Protein G-coated magnetic beads in lanes 6 and 7 and probed with anti-DYKDDDDK (1/1000). Mock-transfected HeLa in Lane 1. A 1/40th part of the lysate was loaded in lanes 2 and 3 before IP and in lanes 4 an 5 after IP. Primary incubations were for 1 hour. Detected by chemiluminescence. Data obtained from Dr P Grozdanov, Texas Tech University Health Sciences Center, USA. ABIN1686783 (2 µg) used to pull down CSTF2 (64 kDa) from Mouse Brain (lane 5) lysate (500 µg protein) while it cannot pull down CSTF2 (64 kDa) from Mouse Liver (lane 6)  lysate (2 µg) protein) using Protein G-coated magnetic beads. A 1/40th part of the lysates were loaded in lanes 1 (Brain) and 2 (Liver) before IP and in lanes 3 (Brain) and 4 (Liver) after IP. Western blot probed with mouse anti-CSTF2 (64 kDa)/CSTF2 (64 kDa) antibody and  detected by chemiluminescence. Cow Rabbit Un-conjugated IC, IF, IP, IHC, WB Immunofluorescent analysis of CstF-64 staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue). Immunohistochemical analysis of CstF-64 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. 200 μL 13 to 14 Days
