High levels of IP6K3 mRNA were found in myotubes and muscle tissues. In myotubes, IP6K3 was upregulated by dexamethasone treatment, which is known to inhibit glucose metabolism.

Ip6k3(-/-) mice had lower blood glucose, less insulin (show INS Antibodies), decreased fat, lower weight, increased plasma lactate, enhanced glucose tolerance, lower glucose during an insulin (show INS Antibodies) tolerance test, and reduced muscle Pdk4 (show PDK4 Antibodies) expression. Ip6k3 deletion extended lifespan.
IP6K3 is highly expressed in cerebellar Purkinje cells. IP6K3-deleted mice display defects of motor learning and coordination. IP6K3-null mice manifest aberrations of Purkinje cells with a diminished number of synapses.
This gene encodes a protein that belongs to the inositol phosphokinase (IPK) family. This protein is likely responsible for the conversion of inositol hexakisphosphate (InsP6) to diphosphoinositol pentakisphosphate (InsP7/PP-InsP5). It may also convert 1,3,4,5,6-pentakisphosphate (InsP5) to PP-InsP4. Alternative splicing results in multiple transcript variants encoding the same protein.
