targeting SmgGDS (show RAP1GDS1 ELISA Kits) induces nucleolar stress, resulting in profound loss of DREAM target gene expression required for G1, S and G2 progression, and ultimately resulting in cell cycle arrest.
Disruption of DREAM and RB-E2F (show E2F1 ELISA Kits) complexes by oncoproteins from DNA tumor viruses leads to upregulation of cell cycle genes and impairs growth-inhibiting pathways, including the p53 (show TP53 ELISA Kits)-mediated downregulation of cell cycle genes. [review]
This study shows the application of hydrogen-deuterium exchange mass spectrometry and site-directed mutagenesis to investigate the Ca(2 (show CA2 ELISA Kits)+) binding properties and the subsequent conformational changes of full-length DREAM. EF-hands undergo large conformation changes upon calcium binding even though the EF-1 hand is not capable of binding to Ca(2 (show CA2 ELISA Kits)+).
Our approach yields high confidence ranked target gene maps for TP53 (show TP53 ELISA Kits), DREAM, MMB-FOXM1 (show FOXM1 ELISA Kits) and RB-E2F (show E2F1 ELISA Kits) and enables prediction and distinction of CC regulation. A web-based atlas at www.targetgenereg.org enables assessing the regulation of any human gene of interest.
downregulation of these genes through the p53 (show TP53 ELISA Kits)-p21 (show CDKN1A ELISA Kits)-DREAM-CDE/CHR pathway appears to be a principal mechanism for G2/M cell cycle arrest by p53 (show TP53 ELISA Kits).
Our results support the hypothesis that KChIPs enhances Kv4.2 functional expression by a 1 : 1 suppression of the N-terminal FERN domain and by producing additional positive regulatory effects on functional channel expression.
GCM1-directed villous trophoblast differentiation is repressed by DREAM
p53 (show TP53 ELISA Kits) can repress transcription of cell cycle genes through a p21(WAF1/CIP1 (show CDKN1A ELISA Kits))-dependent switch from MMB to DREAM protein complex binding at CHR promoter elements.
The potassium channel interacting protein 3 (DREAM/KChIP3) heterodimerizes with and regulates calmodulin function.
expression of calsenilin leads to a disruption of presenilin 1 (show PSEN1 ELISA Kits)/gamma-secretase-mediated epsilon-cleavage of N-cadherin (show CDH2 ELISA Kits), which results in the significant accumulation of N-cadherin (show CDH2 ELISA Kits) C-terminal fragment 1
Genome-wide analysis of trigeminal neurons in daDREAM transgenic mice identified cathepsin L (show CTSL1 ELISA Kits) and the monoglyceride lipase (show MGLL ELISA Kits) as two new DREAM transcriptional targets related to pain.
association of PS1 (show PSEN1 ELISA Kits) carboxyl peptide (residues 445-467, HL9 (show DDC ELISA Kits)) with DREAM is calcium dependent and stabilized by a cluster of three aromatic residues: F462 and F465 from PS1 (show PSEN1 ELISA Kits) and F252 from DREAM.
The association of calcium-bound calmodulin (CaM) with DREAM is mediated by a short amphipathic amino acid sequence located between residues 29 and 44 on DREAM.
The protein DREAM decreases development of L-DOPA-induced dyskinesia in mice and reduces L-DOPA-induced expression of FosB (show FOSB ELISA Kits), phosphoacetylated histone H3 (show HIST3H3 ELISA Kits), and dynorphin-B in the striatum
NS5806 directly interacts with KChIP3 and modulates the interactions between this calcium-binding protein and the T1 domain of the Kv4.3 (show KCND3 ELISA Kits) channels through reorientation of helix 10 on KChIP3
studies define the critical opposing functions of DREAM and USF1 (show USF1 ELISA Kits) in inhibiting and inducing A20 (show TNFAIP3 ELISA Kits) expression
Expression of DREAM in neuroblastoma cells enhances cisplatin mediated caspase-3 (show CASP3 ELISA Kits) activity.
Restoring CANT1 (show CANT1 ELISA Kits) levels in neuroblastoma clones recovered the phenotype, thus confirming a key role of CANT1 (show CANT1 ELISA Kits), and of the regulation of its gene by DREAM, in the control of protein synthesis and degradation
In cardiomyocytes, [Ca2 (show CA2 ELISA Kits)+]i-activated calmodulin kinase II (CaMKII (show CAMK2 ELISA Kits)) activates downstreamregulatory element (DRE) binding transcription factor DREAM, which consequently suppresses the expression of L-type calcium channel 1C-subunit gene (Cacna1c (show CACNA1C ELISA Kits)).
zebrafish calsenilin is involved in endocrine cell differentiation and morphogenesis within the pancreas
This gene encodes a member of the family of voltage-gated potassium (Kv) channel-interacting proteins, which belong to the recoverin branch of the EF-hand superfamily. Members of this family are small calcium binding proteins containing EF-hand-like domains. They are integral subunit components of native Kv4 channel complexes that may regulate A-type currents, and hence neuronal excitability, in response to changes in intracellular calcium. The encoded protein also functions as a calcium-regulated transcriptional repressor, and interacts with presenilins. Alternatively spliced transcript variants encoding different isoforms have been described.
