    Specificity

    Clone VI F11 is useful for staining rat and human surfactant protein D . Antigen Distribution on Tissues and Cells: Alveolar type II cells, alveolar macrophages, two types of Clara cells SP-A+ and SP-A-, and extracellular deposits. Alveolar type III cells stain negative. No reaction on Rat skin, colon, kidney and liver.

  • Application Notes

    Immunohistochemistry on Frozen Sections: 0.5 μg/mL (1/800) for Rat Cryosections and10-20 μg/mL (1/20-1/40) for Human Frozen Sections. Immunohistochemistry on Paraffin Sections: 10-20 μg/mL (1/20-1/40) for Rat paraffinsections (microwave pretreatment for antigen retrieval is recommended). Suggested Positive Control: Rat lung. Has been described to work in ELISA and Western blots.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Surfactant protein D is a Ca2+ -dependent carbohydrate binding protein with a similar structure as other C-type mammalian lectins such as conglutinin and SP-A. The molecular weight is 620 kD under non reducing conditions and 43 kD under reducing conditions. The function of SP-D is not clearly defined. SP-D enhances the production of oxygen radicals in alveolar macrophages and regulates some actions of SP-A, which is the most abundant surfactant protein.
