Application Notes: Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhIP-10 (100.0 ng/mL), a concentration of 2-3 μg/mL of this antibody is required. ELISA: To detect hIP-10 by direct ELISA (using 100 l/well antibody solution) aconcentration of at least 0.5 μg/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with compatible secondary reagents, allows the detectionof 0.2-0.4 ng/well of recombinant hIP-10. Western Blot: To detect hIP-10 by Western Blot analysis this antibody can be used at aconcentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hIP-10 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.

Background: Interferon-gamma-inducible 10 kD protein (IP-10), is a CXC chemokine with chemoattractant properties for CD4-positive T cells and inhibits early normal and leukemic hemopoietic progenitor proliferation. IP-10 is produced by a wide variety of cell types ranging from neutrophils and monocytes to hepatocytes, endothelial cells and keratinocytes. The cytokine is reported to be involved in a scala of inflammatory pathologies such as HIV encephalitis, cutaneous T cell lymphoma, chronic hepatitis and acute anterior uveitis.
