Application Notes
- For flow cytometry dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. For functional studies in vitro dilutions have to be made according to the amounts of CD96 to be inactivated.

Background
- The monoclonal antibody NK92.39 reacts with CD96, also known as T cell-Activated Increased Late Expression (Tactile). CD96 promotes natural killer (NK) cell adhesion to target cells expressing the poliovirus receptor (PVR/CD155). Furthermore, CD96 stimulates cytotoxicity of activated NK cells, and mediates acquisition of PVR from target cells. NK cells have due to the receptor CD96 a dual receptor system that recognizes nectins and nectin-like molecules on target cells. This mediates NK cell adhesion and triggering of effector functions. As PVR is highly expressed in certain tumors, this receptor system may be critical for NK cell recognition of tumors. The monoclonal antibody NK92.39 blocks binding of soluble poliovirus receptor (PVR) to NK92 cells.
