Antibody to Human Antithrombin III. This Antithrombin III antibody gives a single arc when tested by IEP against human plasma. Identity is confirmed by double diffusion (Ouchterlony) against human plasma and a known anti-human antithrombin III. The cross reactivity of this product against the sera of the species listed below has been assessed by double diffusion using 10 μL of antisera against 5 μL of species sera. These results are intended as a guideline only.

Human Antithrombin III purified from plasma, shown to be homogeneous by SDS-PAGE.
RID: 6l antiserum / cm2 gel vs.5 μL neat - 1/10 dilution human plasma. Double Diffusion: 10l antiserum vs.5l plasma. IEP: 100l antiserum vs.5l plasma. The use of 3 % PEG 6000 with 1.2 % agarose in a suitable buffer (such as TBE or Tris-barbital pH >8.2) is recommended. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Background
Antithrombin III (ATH III) is a plasma protein synthesised in the liver. Normal plasma levels are 115 to 160 mg/L. ATH III consists of a single polypeptide chain and has a molecular weight of 58 kDa. ATH III is a serine protease inhibitor and regulates coagulation by the formation of covalently liked complexes. ATH III also has a heparin binding site and in the presence of heparin the inhibition of thrombin, factors IXa, Xa and XIa is enhanced 1000 fold. Low levels of ATH III (functional or concentration) are associated with deep vein thrombosis and pulmonary embolism.
