The antiserum does not cross react with any other component of human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. of this antiserum has not been tested in detail, however in double radial immunodiffusion a reaction has been found with Rhesus monkey, Cynomolgus, Baboon and Chimpanzee, no reaction with mouse.

Purification
Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other plasma proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
Immunogen
Alpha-1 antitrypsin is a serine protease inhibitor. It has a molecular weight of 54,000, its mean concentration in normal serum is 2.5 mg/ml. The highly purified immunogen is isolated from pooled normal plasma. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Application Notes
In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of alpha-1antitrypsin in human plasma or other body fluids or to determine its concentration. The presence of non precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive non-precipitating antibody-binding techniques if proper controls are included.

The reactivity of the antiserum is restricted to alpha-1antitrypsin. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified alpha-1antitrypsin.
