    Tetramethylrhodamine (TAMRA), a fluorescent dye, has gained significant popularity for its application in the labeling of oligonucleotides. Originally, due to its susceptibility to ammonium hydroxide, TAMRA NHS Ester was initially employed to label amino-modified oligonucleotides. However, subsequent research revealed that TAMRA displays resilience against UltraMild deprotection and hindered amines, such as t-butylamine. This discovery led to the swift introduction of supports and phosphoramidites containing TAMRA. The utilization of TAMRA-modified oligonucleotides has played a crucial role in both Förster resonance energy transfer (FRET) and real-time PCR applications.
