Target Description: SH3 and multiple ankyrin repeat domains protein 1, or shank 1 is a member of the shank family. Shank 1 binding domains include a SH3 domain, a proline rich domain, a PDZ domain and ankyrin repeat domains. Unlike other shank family proteins, shank 1 is found in only in brain in the neuropil of cortex, CA1 region hippocampus and molecular layer of cerebellum, amygdala, substantia nigra and thalamus. Cellularly, shank 1 can be found in cytoplasm, cell junctions, postsynaptic cell membrane and postsynaptic density. Many isoforms of shank 1 exist, and some are expressed during development, while others are expressed in mature tissue. Shank 1 acts as a scaffold protein anchoring and connecting membrane bound receptors and other proteins to the cytoskeleton in neurons to enable cell signaling. In this manner, Shank 1 may play a role in synapse formation and dendritic spine maturation. Mutations in this gene have been associated with autism spectrm disorder and diabetic encephalopathy.
