Background

Synonyms: SH3 and multiple ankyrin repeat domains protein 2 (Shank2) (Cortactin-binding protein 1) (CortBP1) (GKAP/SAPAP-interacting protein) (Proline-rich synapse-associated protein 1) (ProSAP1) (SPANK-3)
Target Description: SH3 and multiple ankyrin repeat domains protein 2, or shank 2 is a member of the shank family. Shank 2 binding domains include a SH3 domain, a proline rich domain, and a PDZ domain. Shank 2 is found in kidney, liver tissue, and epithelial cells. In brain, Shank 2 is expressed at the apical cell membrane, cytoplasm, synapse, postsynaptic density, growth cone, and dendritic spine. Some isoforms of shank 2 are expressed during development, while others are expressed in mature tissue. Shank 2 acts as a scaffold protein anchoring and connecting membrane bound receptors, including NMDA and glutamate receptors and other proteins to the cytoskeleton in neurons to enable cell signaling. In this manner, Shank 2 may play a role in synapse formation and dendritic spine maturation. Mutations in the Shank 2 gene have been associated with autism spectrm disorder and blount disease.
