    Assay Procedure

    Highly purified Scl70 is bound to microwells. Antibodies to this antigen, if present in diluted serum, bind in the microwells. Washing of the microwells removes unbound serum antibodies. Horseradish peroxidase (HRP) conjugated antihuman IgG immunologically bind to the bound patient antibodies forming a conjugate/antibody/antigen complex. Washing of the microwells removes unbound conjugate. An enzyme substrate in the presence of bound conjugate hydrolyzes to form a blue color. The addition of an acid stops the reaction forming a yellow endproduct. The intensity of this yellow color is measured photometrically at 450 nm. The amount of colour is directly proportional to the concentration of IgG antibodies present in the original sample.

    Background

    Anti-Scl70 (also called anti topoisomerase I after the type I topoisomerase target) is an anti-topoisomerase antibody type of antinuclear autoantibodies, seen mainly in diffuse systemic scleroderma (with a sensitivity of 28-70 %), but it is also seen in 10-10 % of cases of the more limited form of systemic scleroderma called CREST syndrome Anti Scl70 antibodies are associated with more severe scleroderma disease. The etymology of scl70 consists of an abbreviation of scleroderma and a 70 kD extractable immunoreactive fragment that can be obtained from the otherwise larger (100-105 kD) target topoisomerase antigen (called the SCL70 Antigen) of the antibodies.
