Background
- KMT5A, also known as N-lysine methyltransferase KMT5A isoform 1, is an enzyme which catalyzes mono-methylation of histone H4 at lysine 20 (H4K20me1). It is implicated in cell-cycle-dependent transcriptional silencing and mitotic regulation in metazoans. It acts as a barrier to prevent cellular senescence through chromatin-mediated regulation of senescence-associated metabolic remodeling. It mediates monomethylation of p53/TP53 at 'Lys-382', leading to repress p53/TP53-target genes. The loss of KMT5A concurrently stimulate nucleolar function and retinoblastoma protein-mediated mitochondrial metabolism. Recombinant human KMT5A, fused to His-tag at C-terminus, was expressed in insect cell and purified by using conventional chromatography techniques.
