HPL – Questions and Answers (FAQ)

What is the difference between ELAREM™ Perform and ELAREM™ Perform FD PLUS?

ELAREM™ Perform is the standard research-grade variant with a natural fibrinogen content and requires the use of an anticoagulant. ELAREM™ Perform FD PLUS is fibrinogen-depleted, prevents clotting without additives, and is therefore better suited for sensitive applications and 3D models.

Do I need an anticoagulant for ELAREM™ HPL?

Perform: Yes, an anticoagulant is recommended to prevent clotting in the medium. Perform FD PLUS: No, this variant is clot-free and can be used without anticoagulant.

Which cell types are compatible with ELAREM™ HPL?

HPL supports the expansion of many human cell types, including:

  • mesenchymal stem cells (MSCs)
  • fibroblasts, endothelial cells
  • various primary human cells
  • immune cells (depending on protocol)

The optimal concentration should be validated for each specific cell type.

What working concentration is recommended?

Typical concentrations range from 5–10% (v/v). Some applications use 2–20%, depending on the cell type and desired proliferation.

Is HPL a complete replacement for FBS?

Yes, for many applications HPL is a xeno-free, fully human alternative to FBS. However, users should apply a short adaptation protocol when switching from FBS to HPL.

Can I use HPL for 3D cultures or spheroid systems?

Yes. ELAREM™ Perform FD PLUS in particular is suitable for 3D cultures, biofabrication, and hydrogel applications due to its low fibrinogen content and clot-free properties.

Why is fibrinogen-depleted HPL advantageous for certain applications?

Fibrinogen can polymerize in media and cause clot formation. FD PLUS avoids clotting entirely and enables more homogeneous handling in 3D systems, microfluidics, and sensitive cell models.

How do HPL batches differ from each other?

HPL is a natural product, but PL BioScience uses large donor pools to strongly minimize batch-to-batch variability. The products are considered highly standardized.

Can HPL influence differentiation processes?

Yes, due to its natural growth factor profile, HPL can modulate proliferative and differentiation signals. For differentiation-focused applications, the concentration should be adjusted accordingly.
