Based on its differential expression, neuroanatomical distribution and colocalisation, we hypothesise that rat Asb-4 is a gene involved in energy homeostasis.
Expression of imprinted gene Asb4 was unregulated in haploid androgenetic/gynogenetic blastocysts.
In functional endothelial co-culture assays, JAR (show MYO6 Proteins) cells ectopically expressing ASB4 increased endothelial cell turnover and stabilized endothelial tube formation, both of which are hallmarks of vascular differentiation within the placenta.
The interaction of Asb-4 with IRS4 (show IRS4 Proteins) in cell lines mediates the degradation of IRS4 (show IRS4 Proteins) and decreases insulin (show INS Proteins) signaling.
Tumor necrosis factor alpha (show TNF Proteins) upregulates ankyrin repeat and suppressor of cytokine signalling (show SOCS1 Proteins) box protein 4 expression.
Asb-4 is a key regulatory protein in the central nervous system, involved in the control of feeding behavior and metabolic rate.
Asb4 has been identified as an imprinted gene by high-throughput screening using RIKEN cDNA microarray.
The present study demonstrates that Asb-4 interacts with GPS1 (show GPS1 Proteins) and inhibits JNK (show MAPK8 Proteins) activity.
These results suggest that ASB-4 plays pivotal roles in mammalian testis development and spermatogenesis.
The protein encoded by this gene is a member of the ankyrin repeat and SOCS box-containing (ASB) family of proteins. They contain ankyrin repeat sequence and SOCS box domain. The SOCS box serves to couple suppressor of cytokine signalling (SOCS) proteins and their binding partners with the elongin B and C complex, possibly targeting them for degradation. Multiple alternatively spliced transcript variants have been described for this gene but some of the full length sequences are not known.
