  • Application Notes
    Approved: ELISA (0.5 - 1 μg/mL), Flo (25 μg/mL), IHC, IHC-P, WB (1 - 2 μg/mL)

    Usage: Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and ELISA. Western Blot: 1-2 μg/mL. Detection limit is approximately 25 ng/lane under non-reducing conditions. Reducing conditions are not recommended. Flow Cytometry: 25 μg/mL. For intracellular staining, cells must be fixed and permeabilized using 4 % paraformaldehyde and 0.1 % saponin in PBS. Add 10 μL of the diluted solution to 1-5x10^5 cells in a total reaction volume not exceeding 200 μL. Incubate for 30 minutes and then wash cells with 0.1 % saponin prior to addition of a secondary developing reagent. Cells should be washed a final time in 0.1 % saponin prior to flow cytometric analysis. Direct ELISA: 0.5-1 μg/mL. Detection limit is ~5 ng/well. Immunohistochemistry: 8-25 μg/mL with appropriate secondary reagents to detect IL-17D in paraffin-embedded normal heart tissue sections.
