Description
Gli-3 (also known as Zinc Finger Protein Gli-3 or GLI-Kruppel family member GLI-3) belongsto the GLI C2H2-type zinc-finger protein family and contains 5 C2H2-type zinc fingers. Gli-3is very important for normal limb and brain development and is implicated in thetransduction of Shh signal. Gli-3 is a nuclear protein expressed in a wide variety of normaladult tissues, including lung, colon, spleen, placenta, testis, and myometrium. Defects inGli-3 are the cause of Greig cephalo-poly-syndactyly syndrome (GCPS), an autosomaldominant disorder-affecting limb and cranio-facial development. Two isoforms of humanGli-3 have been reported. One is the full-length protein at ~170-190kDa and the other is atruncated isoform at ~80kDa.

Application Notes
ELISA (1/6000-1/30000). Western Blot (1/500-1/2000). Immunohistochemistry on Paraffin Sections (1.25 µg/ml). This antibody was validated for use in immunohistochemistry on a panel of 21formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigenretrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides wereincubated with biotinylated secondary antibody LS-D1, followed by alkalinephosphatase-streptavidin and chromogen. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
