    Purification

Ammonium Sulphate Precipitation followed by Antigen Affinity Chromatography using the immunizing peptide

    Immunogen

Synthetic peptide from the internal region of the protein sequence according to NP_002713.1. Genename: PPY
  • Application Notes

Peptide ELISA: Antibody Detection Limit Dilution: 1/32000. Western Blot: Preliminary experiments gave an approx 35-38 kDa band in human pancreaslysates after 1 μg/mL antibody staining. Please note that currently we cannot find anexplanation in the literature for the band we observe given the calculated size of 10.8 kDaaccording to NP_002713.1. The 38 kDa band was successfully blocked by incubation withthe immunizing peptide. Immunohistochemistry: 3 μg/mL. In paraffin embedded human pancreas, shows strong cytoplasmic staining of rare cellslocated at the periphery of islets of Langerhans.
    Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.

  • Target

    PPY (Pancreatic Polypeptide (PPY))

    Alternative Name

    Pancreatic Prohormone

    Background

Pancreatic hormone (PPY) is synthesized in pancreatic islets of Langerhans and acts as a regulator of pancreatic and gastrointestinal functions and may be important in the regulation of food intake. Plasma levels of this hormone have been shown to be reduced in conditions associated with increased food intake and elevated in anorexia nervosa. In addition, infusion of this hormone in obese rodents has shown to decrease weight gain.Synonyms: Obinepitide, PNP, PPY, Pancreatic polypeptide
