  • Application Notes

    ELISA: (Direct): To detect mGM-CSF by direct ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant mGM-CSF. (Sandwich): To detect mGM-CSF by sandwich ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with Polyclonal Anti-Murine GM-CSF (PP003P1 or PP003P2) as acapture antibody, allows the detection of at least 0.2-0.4 ng/well of recombinantmGM-CSF. Western Blot: To detect mGM-CSF by Western Blot analysis this antibody can be used at a concentrationof 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detectionlimit for recombinant mGM-CSF is 1.5-3.0 ng/lane, under either reducing or non-reducingconditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Store the antibody at 2-8 °C prior to restoration. Restore with 1.0 ml of deionized water (or equivalent). For extended storage add glycerol to 50% and then aliquot contents and freeze at -20° C or below. Centrifuge product if not completely clear after standing at room temperature. This product is stable for one month at 2-8 °C as an undiluted liquid.

    Background

    The cytokine GM-CSF stimulates the growth and differentiation of hematopoietic precursor cells from various lineages, including granulocytes, macrophages, eosinophils and erythrocytes. Used in myeloid reconstitution following bone marrow transplant, bone marrow transplant engraftment failure or delay, mobilization and following transplantation of autologous peripheral blood progenitor cells, and following induction chemotherapy in older adults with acute myelogenous leukemia.Synonyms: CSF2, Colony stimulating factor 2, GMCSF, Granulocyte-macrophage colony-stimulating factor, Molgramostin, Sargramostim
