    Application Note: Anti-Elastase Biotin Conjugated Antibody has been tested by ELISA and western blot. This product is assayed against 1.0 μg of Elastase in a standard capture ELISA using Peroxidase Conjugated Streptavidin #S000-03 and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) code # ABTS-100 as a substrate for 30 minutes at room temperature.  A working dilution of 1:4,000 to 1:20,000 of the reconstitution concentration is suggested.

    Western Blot Dilution: 1:500 - 1:2,000

    ELISA Dilution: 1:5,000 - 1:20,000

    Background

    Background: Anti-Elastase antibody detects the Elastase protein. Elastase metabolizes elastin, an elastic fiber, which together with collagen, determines the mechanical properties of connective tissue. The neutrophil form breaks down the Outer membrane protein A (OmpA) of E. coli and other Gram-negative bacteria. Elastase also has the important immunological role of breaking down Shigella virulence factors. This is accomplished through the cleavage of peptide bonds in the target proteins. The specific peptide bonds cleaved are those on the carboxy side of small, hydrophobic amino acids such as glycine, alanine, and valine. This antibody is suitable for researchers investigating biochemistry and stem cells.
