Specificity
- Reacts with chondroitin sulfate proteoglycan from cartilage (aggrecan). Species Reactivity: Reacts with human. No crossreactivity with bovine nasal cartilage, rat chondrosarcoma, chick or shark cartilage aggrecan. Other species not tested. Controls: Purified antigen, cartilage extracts or mature cultured chondrocytes.

Application Notes
- ELISA: 1:500-1:1,000 (highest ELISA reactivity of our 3 anti-chondroitin/keratan sufate proteoglycans). In solid phase immunoassays, endo-beta-galactosidase digestion of aggrecan increases the immunoreactivity of the antibody. Also reacts with papain-digested aggrecan. Immunohistochemistry: (paraffin embedded): 1:50-1:3,000. Treat tissue sections with 2 mg/ml testicular hyaluronidase in 50 mM Na-acetate buffer, pH 5.5 for 1 hr at 37C, followed by treatment with 0.2 mg/ml Protease XXIV in TBS, pH 7.2, for the same time and temperature. Immunoprecipitation: 1:50-1:300 Western blotting: 1:100-1:300 Reacts with 3 trypsin-generated fragments of human cartilage aggrecan (prior digested with chondroitinase and endo-beta-galactosidase) which have apparent MW of 55, 65 and 95 kDa. The optimal dilution for a specific application should be determined by the researcher.
