    Specificity

    The amino acid sequence (SPLYSYTTDGNDTVT) used as immunogen for this is 100 % homologous in human, 93 % homologous in vervet/green monkey and 80 % homologous in chimp. Isoforms of different molecular weights have been described for TIM-1. The sequence used for immunogen is conserved between TIM-1 (364aa, NP_036338) and TIM-1 isoform CRA_a (194aa, EAW61615.1). Therefore, the molecular weight observed in western blots may vary depending on the forms present.

    A synthetic peptide (SPLYSYTTDGNDTVT) of human HAVCR1/TIM-1protein were used as immunogen. This sequence corresponds to amino acids (aa) 248-262 of NP_036338 and aa 84-98 of EAW61615.1. Isoforms of HAVCR1/TIM-1 have been described. For example, NP_036338 is 364 aa and EAW61615.1 is 194 aa. EAW61615.1 is referred to as isoform CRA_a. Percent identity by BLAST analysis: Human, Gorilla (100%), Monkey (93%), Dog (80%).

    Usage: Immunohistochemistry: This antibody was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in  pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for this antibody was determined to be 2.5-5 μg/mL.
