  • Specificity

    This antibody detects CD8a, the alpha chain of the CD8 antigen. CD8 is expressed on T-cells, and exists either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. The CD8a antibody has been reported to block antigen presentation via MHC class I and inhibit T cell responses to IL-2. This antibody has also been used for depletion of CD8a+ cells.

    Flow Cytometry: Use 10 μL of the suggested working dilution to label 1x10^6 cells in 100 μL. The Fc region of monoclonal antibodies may bind non-specifically to cells expressing lowaffinity Fc receptors. Immunohistochemistry on Frozen Sections. Immunohistochemistry on Paraffin Sections: This antibody has been reported as workingwell on Zinc-fixed paraffin-embedded sections. However, inconsistent results have beenreported on formalin-fixed paraffin-embedded tissue sections. Immunoprecipitation.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell to cell interactions within the immune system. The CD8 antigen, acting as a coreceptor, and the T cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. CD8 is expressed on most thymocytes and approximately 1/3 of peripheral blood T cells in humans. CD8 alpha/beta heterodimers are expressed only on TCR alpha/beta T cells, whereas CD8 alpha homodimers can be expressed on alpha/beta and gamma/delta T cells and some NK cells in humans.
