Application Notes
Immunoblotting: 0.5 μg/mL for HRPO/ECL detection. Recommended blocking buffer: Casein/Tween 20 based blocking and blot incubationbuffer. Included Potitive Control: Cell lysate from untreated MDA-MB 468 cells (See Protocols formore details). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Protocol
Positive Control: Cell lysate from untreated MDA-MB-468 cells, breast adenocarcinoma(Human)Format: Lyophilized cell lysate from serum starved MDA-MB-468 cells. Reconstitution: Restore by addition of 200 µl H2O. After complete solubilization add 200µl 2x SDS-PAGE sample buffer, mix and incubate at 90°C for 5 min. Application: The positive control cell lysate is recommended for immunoblot applications. 20 µl of positive control cell lysate correspond to ca. 20. 000 cells. Use 20 µl/lane (mini gel) for HRPO/ECL detection of the target proteins. Please NOTE: The lyophilized cell lysates conatin SDS and are not recommended forapplications with native proteins such as in immunoprecipitation.

Background
This protein encoded is a cytoplasmic nonreceptor protein kinase which may function as an intracellular signal transducer in epithelial tissues. Overexpression of this gene in mammary epithelial cells leads to sensitization of the cells to epidermal growth factor and results in a partially transformed phenotype. Expression of this gene has been detected at low levels in some breast tumors but not in normal breast tissue. Its presence in the nucleus appears to be linked to suppression of tumor progression.
