Usage: Suitable for use in ELISA, Flow Cytometry and Immunohistochemistry. ELISA: 0.5-1 μg/mL). Immunohistochemistry (frozen): 25 μg/mL detects SPARC on mouse ovary tissue. Flow Cytometry: 25 μg/mL. For intracellular staining, cells must be fixed and permeabilized using 4 % paraformaldehyde and 0.1 % saponin. Dilute this antibody to 25 μg/mL and add 10 μLof this solution to 1-5x10^5 cells in a total reaction volume 200uL. Following a 30 minute incubation, cells should be washed with 0.1 % saponin prior to adding 10 mL of a 25 μg/mL stock solution of a secondary developing reagent such as goat anti-rat IgG conjugated to a fluorochrome. Cells should be washed for a final time in 0.1 % saponin prior to flow cytometric analysis. -
