Application Details

Application Notes: Detection of equine IFN-γ producing cells by flow cytometry. 5 μL is recommended for staining of 1 million cells in a total volume of 50 μL. PF647P is excited by the red laser (633 nm). The excitation max is 654 nm and the emission max is 672 nm. Detection of IFN-γ by flow cytometry in viable equine PBMC. Cells were stimulated for 16 hours in the presence of PMA/ionomycin and Brefeldin A. Cells were then fixed and permeabilized using 4 % paraformaldehyde and saponin, and subsequently stained with MT166- PF647P (solid line). Matched isotype control antibody (dashed line). The histogram derives from gated events of typical lymphocyte characteristics in forward and side light scatter. Flow cytometry was performed on a BD FACSVerse system.
